Short of upgrading your hardware, I'd start by closing some of those icons in your taskbar tray that you don't need (stuff like Quicktime, Winzip, etc. run in the background and eat up extra memory).
Run a scandisk and defrag on your hard drive every few weeks. Scan your PC periodically for viruses and spyware that may be causing your system to run slow.
Right click on My Computer, go to Properties, Advanced, and under Performance click on the Settings button. Set it to "Adjust for Best Performance." This will take away all the extra visual effects (like the blue XP skin on your windows and taskbars) but that'll free up some more resources that could be used elsewhere and makes a noticeable difference on slower machines.
Otherwise, make sure all your programs are within the recommended system requirements of your PC.
Memory is the cheapest bang for your buck hardware upgrade. If you are running Windows XP with 256MB of RAM or less then you might want to consider upgrading to at least 512. It'll make a considerable difference for anything outside of word processors and internet browsing.

If you don't have the budget for the first two suggestions, First of all use the msconfig to disable any unwanted programs from starting up when Windows starts up, next use ad-aware to get rid of any spyware that is on your computer. If you have Norton Anti-Virus or Internet Security replace them, the Norton products are big memory hogs and slow down your system. Then use ccleaner and Toniarts EasyCleaner to dump temp files and invalid registry entries.

If you have a lot of pictures, graphics, and/or video games on your PC, it's going to run slow.
You can do the following to help speed it up a bit:
1.Start
2.Program Files
3.Accessories
4.System Tools
5.Disk Cleanup
Then repeat steps 1-4 and then choose Disk Defragmenter.
Hope this helps!

go to:
Start
Accessories
System Tools

or it can be you local friendly neighborhood Mr. Virus breaking and entering in the back door with his friends Tojan and Worm
spreading chaos throughout your system slowing everything down which is why kids it's always important to have a virus protector.

I have used many types of drive/registry cleaner/fixers, and absolute best one I've (I still use it) is Advanced Uninstaller Pro 2006. It is free for 20 days, then costs 40 dollars if you want to keep it. Use it to clean your computer up and if you really like it, buy it. Defintely worth it.

Run Disk Cleanup, and then Disk Defragmenter twice. Then go into Add/Remove Programs in Control Panel and remove programs you don't want (make sure you know what they are first.)
