Ok I was just looking this up as well, and so far I have found... Queen Victoria Market, The Aquarium, Old Melbourne Gaol, Luna Park, Saint Kilda market (along the esplanade every sunday i think...not sure though), Werribee Open Range Zoo or Melbourne Zoo and the Observation Deck at Rialto Tower.
Or if you wanted to go a bit further out you could always drive along the Great Ocean Road and see the famous 12 (or now 8) apostles, or go to Phillip Island to see the fairy penguins.
Good Luck and Happy Travelling!

Well if your into art galleries and drinkning coffee, Melbourne is your city. Go to federation Square (take ur laptop with wireless hub and get free hi-speed internet) or go to the Melbourne Aquarium, Rialto Towers for a view of Melbourne and the Port Philip Bay. Melbourne is also littered with overpriced posh stuck up cafes if thats your thing. Go and pick on goths and emos at Flinders St station. Spit on joggers from the brige on Flinders St. Or get shitfaced and the many nightclubs :)

Melbourne Zoo, The Exhibition building, Federation Square, Melbourne Aquarium. If you want to go further out field head towards Gippsland Lakes, Tarra Bulga Park, Healsville Sanctuary, if you do decide to go out of Melbourne check where the fires are first with regards to accessibility apart from that enjoy yourself.

G'day,
Rather than repeating what the earlier answerers mentioned, I have provided some links that may also be useful to visit to get better information and planning ahead.
If you are going to stay in or close proximity to the city/CBD area, then use public transport to visit the tourist sites in Melbourne area. After visiting the Melbourne sites, you can then rent a car to visit the sites outside Melbourne (eg. Great Ocean Road, Ballarat/Bendigo, the Grampians, Philip Island etc).
Enjoy your holiday and welcome to Melbourne :)

Check out the rialto tower, a cruise on the river (there is a good one at night that leaves from southbank where you can have dinner on board) crown casino, st kilda (luna park, ackland st) fitzroy gardens, china town is good for a meal, so is hardware lane.
If you want some good ideas go to www.thatsmelbourne.com.au they have a big list of attractions in the city which says the addresses opening times and some costs
