Depends on how you look at it. If you mean capable of attacking us and doing damage, I would go with Al Qaeda. North Korea may have nukes and Iran may be close, but they are too far away to be a threat to us. America has the weaponry available to shoot down any nuke launched at us from either country before it could do damage.
However, Al Qaeda doesn't launch weapons, it personally delivers them in secrecy. Intelligence capability is needed to stop them, and our government has shown it doesn't have the intelligence (pun intended) to handle that too well.
But as far as a threat of destroying us or our way of life, I doubt any of the three are capable at all. America is too big and too far away for the size of the armed forces any of the three could send. The resistance would be far too great for any of the three to make any headway. Even a computer attack, while it would cause huge problems and probably death for a rather large group of people, wouldn't even come close to destroying American infrastructure before it was restored.
The biggest threat in terms of capability (though they don't threaten us as much as Iran, North Korea or Al Qaeda) is probably China. While we have the weaponry to shoot down their nukes (though not all of their weapons, their nukes are too slow for our intercepting weapons), they have a large enough population that if they impressed everyone of their population into service they might have the capability to invade and take a fairly sizable chunk of America before we could stop them.
I still think America is too big for even China, but they could effectively take over more or less the American mainland.

Al-Queda.
North Korea and Iran are tied to geographic locations that can be bombed/occupied/nuked if necessary.
Al-Queda, in addition to be extremely patient, isn't tied to any particular location. As such, they can hide in other countries and be protected by those countries -- even if the country in question has no interest in protecting them.

N. Korea is only dangerous because of who they might sell their technology to, and that is likely Al Q if they can pony up the cash. Kim would sell his grandmother if the price was right.
I still really think things can be worked out with Iran if we stop being such idiots about it.
