If used as a plural and in context indicating slang usage, it refers to women's breasts and usually fairly large ones (Hooters, Hooters Girls, "She's got great hooters!").
If used as a singular and in context indicating slang usage, it refers to oral sex performed on a male and this usage usually carries the connotation of "by a female" ("I need a hooter really bad man! I hope Ashley's a good girl tonight..."). This usage is losing out as Hooters spreads the connotation of breasts.
Of course, it certainly has dictionary meanings, but no one is likely to "take back" the word and make it in any way likely someone means something other than the above. Today it is a purely sexual slang term.

Depends on the context exactly. A hooter can be a loud horn, which has also spawned its own verb "to hoot", meaning to blow a horn.
An owl is often referred to as a hooter due to the noise they make.
And of course hooters (the plural) almost always means female breasts.

If your talking about the restaraunt it's a roundabout way of talking about the breasts. Owls have big eyes like how a man would stare at big breast. Consequently Hooters had to start hiring women other than Large breasted women. It didn't hurt them as Men who gawk at women don't mind diversity.
