If you are after night life you try Mykonos or Santorini
If you are after nice beaches you try Cyclades
If you are after landscapes try East Aegean
If you want to combine all in one, then try Crete!

Imagine you come back from your long expected holiday to one of the beautiful Greek Islands.. and the only thing you can say back home is..'it was absolutly a beautiful island', then you have lived/seen little or nothing.
If you go for the Isle of Crete, you can say same adding that you have experienced Greek spirit & hospitality away from the main tourist beaches and sites..
A lot of the answer on your question depends on the time you are able/willing to stay... the Peloponnese (considering that it is an Island due to the Corinthe canal), if you have time to spend, is probably the best choice..
My answers are giving you little guidance; I know, but whatever choice you make, you will go back to Greece.. like me myself over 14 years know..

Islands are islands, but Santorini is different than the rest because of its volcanic past. You could take a very brief boat ride to the small island nearby and walk on soil left behind by volcanic activity from years past, all while you look across the water and admire the view of the bigger island.

Depends on what you're looking for and how long you'll be staying. Santorini has fantastic views and is fun to be for a couple of days, but I think I'd get sick of it pretty quickly; however, if you're looking for natural beauty, that's the place to be. Mykonos is, as previously mentioned, noted for having lots of clubs and parties. Crete is never a bad choice... I loved Chania. Also, if you're looking for any historical/archaeological sites, Crete is your best chance of the islands with Knossos, Phaistos, etc. I also traveled to Tinos on my journey, but didn't find anything particularly special about it, unless you're especially interested in Greek Orthodoxy (It's home to the Church of the Anunciation). Unfortunately, I didn't get to travel to any other islands, but I hear Corfu and Rhodes are incredible as well.

look my man if you're searching for party and fun GO TO MYKONOS. That is # 1 party island in Greece, let alone top 5 in europe. its great. its like paradise there. you will experience things there that you never thought of and would never see anywhere else in the world. from clubs to beaches, its all day all nite party experience with wonderful people. (just watch out for gays!!!lol)
on the other hand if you're looking sightseeing, nightlife, ancient scenery, beaches, basically all in one GO TO SANTORINI!
It's a beautiful island there and everybody there is so kind. there is shopping great food as anywhere in greece, and its just an all round gorgeous island and not to mention the sea cast at the top of the mountains is unbelievable.

Corfu. Its a place you will never forget. It has everything all the islands of Greece have, incuding parties, great beaches, wild nature, archeological (the most fortified medieval city - over 10 castles in the island) all combined with a post romantic Venettian feel and of course the unique music of Corfu. Even Normal Orthodox hymns sound different in Corfu. Italian and British occupation left their mark in everything, from dialect to architecture to music.

It depends what you are looking for. Santorini is a unique landscape and architectural setting, but quite touristic in the town, with many bars and young tourists. Mykonos is beautiful, but it is expensive and touristic, not like Santorini, with higher income tourism but much clubbing, models, gay fashion designers, etc.I personnally love Naxos, it has wonderful beaches and monuments, good food, fun but also quiet. Siphnos and Milos are also lovely, for family-style holidays, but they are picturesque and Milos especially has great beaches. Syros has a neoclassical town, quite different from the rest of the islands, more urban. Rhodos and Kos are big islands, with great beaches, many monuments, but again with toursim focusing on clubbing and so on. Of course, you don't have to do that if you don't like, I have been to all of these places and had a great time, doing the things I chose. You can combine 2 islands. Naxos-Santorini, Syros-Mykonos, Sifnos-Milos, or Rhodos-Kos, these pairs are easy to combine because they are on the same ferry line.

Santorini-but not in August. Too many people and expensive. Greece has over 3000 islands,about 250 inhabited so there is an island for every need. Depends on what you are looking for. Beaches? Night life? meeting people? sailing? scuba diving?

IOS! It's a great little island, lots of party spirit and beautifual beaches too. It's small and really friendly and great fun, I love it there. Also its only 1 and a half hours by boat to Santorini so you can visit there too, its well worth it. It's cheaper to stay in Ios thought and easier to get around. Just get a room in the village and you can walk everywhere you need to go.

It depends what type of holiday you'd like:
I've only been to Crete & Zante and didn't think they were amazing looking but I hear Kefalonia is although that might be a touch quiet for some people.
trying reading sites with destinations guides like lonely planet and directline holidays, which have got reviews of most (if not all) the islands.

Rhodes is lovely. but you should do a cruise of all the greek islands, it is a week long cruise and does 5 islands.
