Get a new door, there are literaly 10 of thousands of them that fit in junk yards around America.

If you dont want to replace the door, you will need to sand the hole, use bondo to fill it, sand it down, prime and paint it, not just he patch but the whole door because you would be able to see the spot. It may be easier and more economical to take it to a professional. I am thinking if there is one rust spot there are probably more.

The best way is to cut all of the rust out and weld in new metal. Anything else is just a band-aid and won't do much good.

a.) do it the professional way and remove the rust around the whole, take a piece of thin metal and weld it to shut the hole, sand it and repaint the door.

b.) do it the "Pimp-My-Ride" / "Make the car Nice lookinging but Fragile" way and just fill the hole with lead.
