Hi, you can try the following itinerary.
1st day,
Sentosa and Mount Faber
2nd day,
Little India and Chinatown as they happen to be on the same MRT (rail) line so it is convenient to visit them and they are unique to Singapore. And Chinatown has many nice food to try (Maxwell Food Centre) and there is an Sri Mariaman, an Indian Temple nearby you can visit. And if you have time to spare, you can take a direct bus from Chinatown to Bugis Junction for a bit of shopping. A famous Chinese temple (Guanyin Temple), Singapore National Library and Raffles Shopping Arcade is somewhere nearby as well. If more time permits, you can go to the Esplanade and the Merlion Park. (This is going to be very tiring though)
3rd day,
I would suggest going to the Botanical Gardens, Orchid Garden and then travel to Orchard Road for a bit of shopping. If you do not want shopping at Orchard Road, you can travel down to the Zoological Gardens then move to the Night Safari which is unique to Singapore.
4th day
Jurong Birdpark (contains the world's largest man made waterfall, well that is the last I heard of), Xiao Guilin, Kent Ridge Park, all of them found on the western part of Singapore
Hope the information is useful for you. Below are some websites you can visit to find out more information.

You can spend 1 day doing a city tour on your own. Are you traveling by Singapore airlines? If you are,you can go for the SIA hop on bus that will cost be only S$3 if not that cost you S$12 for the whole day. It only runs in the city. Going to places like the civil area, Chinatown, botanic garden, Orchard Road, Little India and Arab Street. The bus comes every half an hour. You can hop on and hop off anything time u want to and its unlimited Time. That bus also goes to Sentosa our resort island. So if you want, you can spend 1 day on city tour.
From more information on Singapore you can check out the Singapore tourism website.

Or once you arrival in Singapore. Visit the Singapore Visitor Centre located at changi airport T1/T2 arrival and departure. The customer officer there is more willing to help you then anyone.

erh... 4 days without a side trip to KL is better.
Day 1 - check in, shopping in orchard rd, night shopping & dining (in expensive local & chinese food) in Chinatown.
Day 2 - tourist spots - Zoo, Bird Park, Night Safari, night shopping & dining (inexpensive indian cuisine) in little india and Mustafa (24hrs shopping mall).
Day 3 - tourist spots - Snow City / botanical garden, Sentosa (must go), shopping & dining at VivoCity (largest mall). chill out at St. James Power Station (Pubs/Clubs).
Day 4 - Last minute shopping, check out, bye bye...
You could take a short day trip to JB / Desaru, rather than KL, on day two if you are not into animals.
