let them nurse on mom for at least 6 weeks, sexing them right at birth is easy if you know what to look for. I used to breed guinea pigs. They don't become mature till they are about 12weeks old, so you don't have to worry for awile yet. http://images.google.ca/imgres?imgurl=http://cavyspirit.com/images/ThreeCutePiggies.jpg&imgrefurl=http://www.cavyspirit.com/sexing.htm&h=78&w=157&sz=3&hl=en&sig2=Q__db2BpLnCzyI3DucjsMQ&start=21&tbnid=vBqagPcSwnsQ8M:&tbnh=48&tbnw=97&ei=OZCtRef0EoeCJafj6YYK&prev=/images%3Fq%3D%2Bsexing%2Bguinea%2Bpigs%26start%3D20%26ndsp%3D20%26svnum%3D10%26hl%3Den%26lr%3D%26sa%3DN the pictures on here should help alot! they even have some for newborn baby guinea pigs.

You can sex them from birth, although it can be difficult.

Do NOT let these babies stay with their mother until they are six weeks old. They need to be separated at 3 weeks. At the age of 3 weeks, the male babies can start coming into puberty, and they will reimpregnate the mother.

Dont wait 6 weeks!! guinea pigs can reproduce at 3 weeks of age, so separate them then. go to www.cavyspirit.com for a great guide to sexing them.
good luck, and congratulations!! :)

you can tell usually as soon as they are born find some sites (that have pictures) and if they are male u need to separate them from mum when they are 4 weeks and put them with the father. they need to stay with their mum for a couple of weeks for milk.
