I personally use:
Ad Aware by Lavasoft.

As a back-up, I also use:
Spybot Search and Destory
once in a while to thoroughly check if Ad Aware missed something (they can't be all perfect).

As added protection, I also use:
Spywareblaster.
It does not clean spyware that's already there but it prevents spyware to infect your computer.

Windows Defender is also okay.

By the way, all these are free... just download at Download.com

One of the first applications built to find and remove adware and spyware, Ad-Aware SE Personal Edition's excellent reputation is well justified. The sky-blue, skinnable interface features five buttons. The first two, Status and Scan, lead to the core function of the application. These buttons initiate a scan of your files for adware components. After scanning is complete, the program presents a summary of results, followed by a list from which you select exactly which components to remove. Right-clicking an individual entry gives some information about the piece of suspected adware, though we would like more details.

Ad-Aware SE can alert you to more malignant forms of malware by separating items into critical and negligible categories. The third button, Ad-Watch, is nonfunctional in the Standard version. The fourth button, Plug-ins, shows you which Ad-Aware plug-ins are installed. The fifth leads to the help files.Ad-Aware SE does an excellent job of quickly finding and removing most adware and spyware components, although you will have to restart and rescan for a seriously infected machine. We were pleased to see an auto-update feature included with the program, keeping Ad-Aware up-to-date with the latest adware components. Ad-Aware SE should be part of your arsenal for keeping your machine free of adware and spyware components.

Definitely Windows Live Onecare Beta. It's a anti-virus, anti-spyware, and firewall all in one. And the best bit is that it's free.

if you read the computer mags, youll see Kapersky, as top rated. Its not free, but good stuff isnt, however, like all proper anti virus/spyware/firewall product, they give a free trial. I use this, and have used all major (and minor) security software, ONLY because, i regularly re format (once a week!) as i like to experiment and "play" with my pc, and the quickest way to re do everything, is simply to reformat. (not good for people who store music, photos, only have 1 pc). But doing so, allows me to have a permanent "free" premium antivirus, as i reformat long before the trial runs out. then, i dowload again, same email if they ask for one...! Major software sites, dont plan on people regularly re formatting, and have no software in place to detect repeat customer details! Its one long freebee.

The most popular anti-spyware programs are Lavasoft's AdAware and Spybot. They are also free.

Perhaps I'm a bit paranoid, but I use three programs:
Ad Aware SE Professional.
Spybot Search and Destroy.
Windows Defender.
Whatever you decide to use, check on a regular basis for new versions and definitions.
