Come clean. Let your boss know that you want to move jobs and give him a good reason (even if it's not true). Something like you want to work closer to home or whatever. Make sure he knows you are getting interviews and when you get an offer. Give enough notice for him to replace you (I think 1 month is standard for companies - or agree a time period).
Once you've given your boss a good reason for leaving, kept him up to date and give him enough notice, there's no reason he should stop you.
Use the same reason you gave your boss to the interviewers who can't understand why you want to leave.
Also, be persistent!!! Obviously you want to get out of your job, but the fact that you already have a job means you are not desperate to move. You still have an income to live off, so getting a job in a months time isn't as big of a deal as if you had no current income.
Just make sure you have all communication with your boss in writing (either on paper or by email). That way if anything happens like he/she goes back on their word, you have proof of the conversations you have had.
If you have a meeting in their office, send them a summary email or letter afterwards and make sure you keep a copy. Emails are better because they are time stamped and you can prove they have been sent. You can CC yourself into your emails so you receive a copy and keep the sent item as well.
By keeping your boss aware of what is going on, you do not need to creep about and worry about him finding out. Making sure you have communication in writing means if anything goes terribly wrong, you will come out of it unscathed. Making sure you come up with a good reason for moving (true or not) means other people will be more inclined to help you move jobs.
And if you're lucky, by letting your boss know you want to leave, you may receive an offer of a pay rise as an incentive to stay (hopefully). Just make sure you keep calm and don't lose your temper or say something you will regret. Even once you have a new job, don't tell your boss where to stick it on your last day. Walk out of the office as the bigger and better man.

Try to be straightforward; tell your employer the matter bothering you plainly, without any sort of exaggeration. Seek his suggestion and help to overcome the situation.
And also in an interview clearly state your situation and try to not blame your employer, as your employer is not present over there.
Try to point out your disadvantage, explain about your efforts to overcome this and expose the reason why your efforts went in vain.
Though it is very difficult, but if you can explain some points which you did not comply in this process that will create an innocent impression. Good luck.

Be positive in the interview. "I work for a great boss but I want to further my career, take on new roles/tasks, want to work for your company as you are a more progressive get ahead modern company with a more vibrant workplace" etc.
If you say something good about your boss and word gets back to him, he won't be so pissed off.
When you've done as many interviews as I have, both as interviewee and interviewer, the bullshit just rolls off your tongue.
