Overall it's the Green Goblin but out of the symbiotes, Venom

The Green Goblin because their rivalry is now completely personal. The Goblin has managed to:
1. Find out Spiderman's identity
2. Infest Harry (his own son/Peter's best friend)
3. Kidnap Gwen Stacey (Peter's first love) and threw her off a bridge.
4. Kidnapped Aunt May and faked her death (using an actor)
5. Kidnapped May Parker (Peter and Mary Jane's daughter) and her where abouts I still don't know.
Those are just naming things of the top of my head and there's still a lot more.

He has many enemies, I think it would either be, Venom or Green Goblin. Venom mainly was Peters Friend, Eddie Brock. He turned Evil and that friendship with them made Spidey vunrable.

Right now I would say Tony Stark is Spider-Man's #1 enemy. After screwing Spider-Man over by manipulating him to de-mask during Civil War, and Spidey's subsequent switching of sides and going on the run, Iron Man has proven to be Spider-Man's current biggest enemy.

Norman Osborn (in or out of the Green Goblin costume)
Drove his son batty enough to attack his best friend, namely Peter...
Had sex with and fathered children with Gwen Stacey, Peter's first girlfriend...
Drove Flash Thompson to drink again and almost got him killed as a result (Peter's high school bully/friend)...
Arranged the fake death of Peter's Aunt May...
AND he is just an all-around bad guy!!!

I'd say he is his own worse enemy. His insecurity and lack of proactivity causes 99% of his troubles.
