The hub for Harajuku fashion is Sundays in Yoyogi Park. thats where all the fashionable teens congregate to show off their unique styles.
For High fashion, you can also check out the Ginza ward, and for the nightlife fashion, there is no better place than Roppongi.
To arrange your travel, I used http://www.jtbusa.com/ as they got me a great price on my ticket, also they had a ton of information on traveling around Japan, helped me obtain the rail pass I used to travel all over the place (which if you are going for fashion, no need to leave Tokyo. They also have tour packages, I didn't use any, but I have heard good things about that.
There is another site that offers a all inclusive tour package, and in April they are having a Gothic Lolita fashion tour which centers around Harajuku, the link is: http://www.popjapantravel.com/
On my vacation, I am not very fond of the very structured tours, so I just flew over with a rough plan, and played it by ear by what I wanted to do each day.
Another must see thing, in my opinion (however it conflicts with Harajuku, so if you are only staying 1 Sunday, do Harajuku) is Akihabara on a Sunday, the main road is closed to cars, and is opened to foot traffic, you see budding starlets singing and dancing in the street, dressed up in very unique ways, it was a very interesting spot, especially when you see hundreds of otaku snapping photos of each person.

I am currently living in the Tokyo area. It is fun. The lifestyle is nice but I just hate the long commute everyday to work.

I'm going sometime in the summer and I'll be in the harajuku area checking out all the fashions. And btw, it's totally appropriate to spell is Tokio. That's how the local tokyoites spell it! I prefer Edo myself. Drop me a line and I'll treat you to a parfait in one of the Maid Cafes~!
