I'd have to say it'd be the ending of American Beauty or the scene where Rocky Balboa is talking to his son

The rope on a cinder block scene in Old School, Funniest scene ever.

When Phoebe Cates Climbs out of the pool on Fast Times is the overall best scene ever in a movie though.

The end of Carlitos Way.

From the moment he visits his lawyer until the end credits. Around 30 minutes which is a non stop chase scene.

Also from the same director, the pram scene in The Untouchables.

In "Jaws", the scene is at night. The 3 guys are sitting around getting drunk and showing off their scars. Robert Shaw, the boat owner, starts talking about the USS Indianapolis sinking during WWII. He quietly describes how the men in the water were picked off, one by one, by the sharks. Without a doubt, the greatest soliloquy in modern cinema! Great question...

There's a scene in "As Good as it Gets" where Jack Nicholson's character pauses in front of a group of people waiting to see the psychotherapist and asks,

"What if this...is as good as it gets?"

Most people see therapist because they very much fear that they'll never know a normal life or contentment, and he just cut to the heart of it and reinforced the doubts they already had.
