The biggest book ever known to man, was entitled in the original Orianish language (from which we derived our terran-based Semitic languages) "'HLN 'DM'" or in English "How to Serve Man." The book was written on the underside of what we thought was the Great Plains, but was found to be a large scroll which had been dropped and over the centuries grew grass on the backside. 'HLN 'DM' is actually a cook book and describes in detail various Orianish recipes for cooking the hominids they found here on earth. One dish, "Stew of mixed humans with Redwoods, White Slate, and Ferrous Soil," stands out for its shear inventiveness and use of diverse regional flora, fauna, and minerals.
A note scribbled in the margins states (in translation) - "Need Vdactian sentient mussels. will only take 2 eons. Finish when we return."
Scholars have calculated this out to mean that the cook went to the next galaxy for ingredients which were unavailable here. According to the notes, the cook will return to start cooking sometime near the end of August 2007."

I'm going to make the assumption that you are looking for the title of a single fiction novel.
I can't prove it with full word counts, but there are several works-- ancient to modern authors-- that create significant volumes of literature. The "Dune" works, by Frank Herbert, are excellent when you feel bored for a couple months. Anne Rice novels generally weigh in around 125,000 words.
The Greeks and Egyptians were known to love literature but with the loss of the Library of Alexandria and the destruction of some manuscripts over time, it's difficult to asses the largest contributions to literature.
The Japanese gave us what is believed to be the first novel, Murasaki Shikibu's, The Tale of Genji. The modern compilation of the work is reported to be 54 chapters and over 1000 pages. If word counts are estimated based on a standard manuscript page, the word count could be between 250,000 and 500,000 words. The true count is not known because chapters are missing. This is also significant because it was written by a woman.
