Easy! Franskton and Peninsula Airport Shuttle - its the little green and white minibus that leaves from outside the international terminal with all the other shuttle buses. They have several drop-off points in St Kilda and will cost about $20 one-way. My family and I use this service often to get to Frankston and Mornington, and even in heavy traffic we've never missed a flight. timetables, fares etc are all on their website, http://www.fapas.com.au/, and you can even pay online in advance to save cash hassles when you arrive in Melbourne.

The airport is not served by Melbourne's public transport system (except for some very indirect and infrequent local buses).
If you take the Skybus, once you get off walk two (full) blocks south along Spencer St to Bourke St and catch the 96 tram from there. The Collins Street tram is 112, not 12; it doesn't go beyond Fitzroy St in St Kilda. The trams stop running at around midnight. Current fare is $3.20.

Well I usually get to St Kilda by tram. So I would say the best way is to get a taxi into the city and get a tram dont there. There are a couple of stops in St Kilda so ur never to far away from everything.
