Recently built new 'puter and first time using ASUS motherboard. Really like it. Is a P4S800D-X. Everything works good on it, but when I shut it off, and go to restart it later, it says that I have to enter BIOS and select proper boot device. And when I go in there, the HDD does not register right away. When it does, it is greyed out and says new device detected, please reboot. So then I do and I gotta go select the HDD to boot from again, then I hit F10 to save and exit, but it never saves, still gotta do all that over again. Any Help please!! Thank You.

It may be possible that the battery on your motherboard is bad thus not allowing the bios to retain the changes you made and booting from the defaults.

After selecting HDD to boot from, go to the Exit Menu, select Exit and Save Changes from menu. When you select this option (ENTER) a Confirmation Window appears, select Yes to save changes and exit.

I tend to agree with zippo, sounds like a CMOS issue. If the computer is not keeping the correct time is one dead give away on a dead CMOS battery.
