Romanticism stresses the importance of the human imagination, particularly in the presence of nature. Wordsworth's "Tintern Abbey" describes the process. Puritanism emphasises the importance of the individual conscience in moral questions. What the individual imagination is to Romanticism, individual conscience is to to the Puritanism. The romantic "reads " nature; the Puritan, at least historically, believed in reading and interpreting the Bible for himself.
Transcendentalism carries the romantic impulse further. Through imaginative perception, the individual can rise above the real (or quotidian) world we live in and help bring about a better one. One of the best exponents of Transcendentalism is Ralph Waldo Emerson. He got into trouble in Boston for not considering the Bible central enough (all Puritans placed it at the very center of their lives). Like other New England intellectuals of his time, his leanings were toward Unitarianism.
While one can make a case that there are realistic elements in Romanticism--the detailed or "empirical" descriptions of nature, especially in Keats--realism is ultimately opposed to or at least very different from Romanticism and Transcendentalism. As the terminology suggests, the realist believes in the real world, the world we are given, and wishes to work through it. In Melville's "Moby Dick," Captian Ahab could be considered a Romantic-Transcendentalist and the passages describing whales at great length realist, although some are mystical.
Another thing they have in common is that all but Puritanism essentially happened in the 19th century. The heyday of Puritanism was the 17th century.
