But the phrase "the eye of the tiger" refers to focusing on the prey as a tiger does. Once a tiger focuses on a particular animal for prey he will not take his eye off of it until he catches it or he can't run anymore.

Besides all the above (Rocky theme song...) it was taken from a quote in the movie )or maybe it was the other way around.... it is a reference to the "game face" you have to put before the fight.... something like look at the opponent and show no fear and all that BS.
