When you are shopping remember a few things.
CPU speed (processor) try to get at least a 3Ghz, if you are unaware of specifics and sales people since this, they will sell you an older model. Try to get it with a gig of memory also and at least a 160 gig hard-drive too. I noticed many of my friends aren't getting any software with brand new systems either. Make sure you get a WINDOWS disk and a system recovery disk AT LEAST.
so remember:
CPU speed at least 3.0 Ghz
1 Gig of Ram
160 Gig hard-drive
Windows software at the least
Go in with a list and ask for the above, minimum, look at the system info they list also. KNOW what you want. It's good to take a computer savvy friend along.
If you are talking brands, there are many different opinions. The only brand I have seen many problems with is Dell.

iMac, hands down.
1, Design. No tower, the whole computer fits in the 2" behind the monitor. Compact keyboard. Ease of assembly. Plug 'n' play.
2. User friendliness. All the tools you need are at your fingertips. Everything is intuitive. No more going to the Start menu to search for what you need.
3. Virus protection. Macs are immune to viruses and spyware. You don't even need Norton.
4. Stability. Mac's OS is Unix-based. Never crashes. Never breaks down. No SNAFUs. No more tearing out your hair at lost files or e-mails. Don't even bother with an extended warranty.
I have used Dells, Acers and others at various jobs I've held and I hate the counterintuitive, clunky MS operating system. You don't know what you are missing until you get comfy with an Apple. I've been using them since 1984 and I wouldn't take a PC as a gift!
Afterthought: for a little extra $$ you get can the whole MS Office suite on your iMac, so you have the best of both worlds!

Pikachu is a AMD fanboy, YES, AMD processes used to be the best bang for your buck, and had the best highest CPU's a year or so ago, but right now the Intel Core 2 Duos are the best processors outt there. Although in a month or so, AMD should release there next CPU which will probably be better.
I do agree with 2GB's of DDR2 ram though and with the video card. I would only suggest a flatscreen if you need the room. believe it or not, a CRT has a higher quality then an LCD screen, and also has a higher resolution for the size of the screen. If you do get a LCD screen, and your into gaming, then make sure to get one with less then 8MS response time or you will see ghosting.

Look at reviews on PC World. This question pops up to often on Yahoo Answers. You have to remember that most end users have used about 1 or 2 computers from a single manufacturer. They are not necessarily product specialists.
Try looking at computers on www.govconnection.com. You can refine by product specifications and by price.
I have extensive experience with the IBM/Lenovo product line and could point you in that direction, but it may not necessarily serve your needs.
I buy desktops almost every week and prices fluctuate.
The best i can offer is just make sure the computer you get has the following:
Intel Core 2 Processor
1GB-2GB RAM
120GB or greater hard disk
16x DVD Burner
Most computers with this configuration will be under $1,000.

I just know I got all these answer beat. For a minimum make sure your computer has: 2 GB of memory, dual core, 64 bit, and 18X DVD (with ram preferably). Like one answer given. Go to pcworld.com and they will show you what the top computers are for the prices. Pcworld will also explain about the chips, motherboard and any other things you may think of. Also check out the Sunday paper if you are in a big city, cuz sometimes you can get some good deals. Spend $1000 and get maybe $300 in rebates back. check both sides though.

Best you can do is to trade your old computer ( Be sure it's working) why spending a thousand dollars for it! if you can buy a NEW ONE. Whether it's a Desktop or a Laptop, it's up to you for your convenience. Buy the following:
Intel Core 2 Processor
1GB - 2GB DDR
120-160 GB Hard Disk (Seagate / Maxtor)

It's hard to answer your question because i don't know what kind of computer you have. As a general rule you should upgrade your ram, cpu and video card first. These 3 things will give your pc an immediate speed boost.

The best computer has to be custom made.
You can get more memory.
You can get good graphic card NVIDIA Geforce 8800 or 7800GT
2 gigs of RAM at least. 3+ etc is no point
AMD processor.
A big flatscreen :)

as was already stated you need to get the "best" computer for "your" needs.
there are so many to choose from most with more bells and whistles than the average computer user will ever need.
you may also want to think about getting a laptop with docking station vs. a desktop
