It would be hard pressed to label any one game the best, considering how many different genres and tastes are out there. Sports titles are the only genre that can be thrown out at they are more of a collection or a series than an individual game. Here is my attempt at a breakdown:

First/Third Person Shooter
-Halo - Xbox (original over Halo 2 due to it's impact and the energy sword debate)
-James Bond 007: Goldeneye - N64
-Doom - PC

RPG
-Final Fantasy VII - Playstation (I still have yet to play a RPG that rivals the fun of the materia system)
-World of Warcraft - PC

Fighting
-Mortal Kombat - Arcade
-Street Fighter II - Arcade

Old School
-Galaga - Arcade
-Centipede - Arcade
-Pacman

Side Scroller:
Super Mario Bros. - NES
Super Mario Bros. 3 - NES

Adventure:
-Myst - PC

Puzzle
-Tetris - Multiplatform

-Other
-Dance Dance Revolution

Lifetime Achievement:
-Pong - Multiplatform

My finalists would be Halo, Final Fantasy VII, Galaga, and Tetris with Tetris getting the nod due to its ease of introduction, lasting appeal, and replayability. It has stood the test of time moreso than any other game and is still being played by bored office workers, distracted trigonometry students, and everyone else to to this day.

If you are talking about arcade games then I must say Geometry Wars or Rampage World Tour. If you are talking about any game, even modern, then I would say the Halo series.

That's a tough question. Probably pac-man or mario bros, considering how those two games impacted the entire industry.
