He's having a throat culture.

How can i stop him from squirming ? Can i sit him on my lap ?

What's best way to restrain him ? Because i know, he'll put up a fight.

It takes all of five seconds. If you want you can place him in your lap . Have him put his arms on his sides (he should be facing the doctor with his back to you) and then put your arms around him. When it's time for the culture hold him firmly so that he thinks you're giving him a big hug. Squeeze and tell him that you love him. Good luck!

I lay my daughter down on the table and hold her head still while the nurses take care of the rest.

Sit him on your lap, with him facing forward. PLace you hands on either side of his face/jaw with your thumbs hooked behind his ears.
