I was taking a look at the other answers to this questions and I saw a lot of mistakes.
First, never end a cover letter with a formula like "Sincerely yours" or "yours truly", or, even worst "Very tryly yours". This is something you can end a love letter with. It's too familiar and intimate. A cover letter should end in "Sincerely, John Doe" and this is all.
The beginning of a cover letter is represented by the addressing formula, which should look like:
Dear John Doe
Dear Sir/Madam
Dear Hiring Manager
or
To whom it may concern
Anything else will sound too familiar or too informal
The content of a cover letter should not say that you have attached a resume and this is all. By no means! Here's what the content of a cover letter should contain (depending on the job you are applying for and the destination):

What else can I say. Good luck and read some secure sources.

A cover letter is for any information you don't put in your resume, and also a way to become more personal with the employer.
So what you write is: why you'd like to work for that specific company, what you can bring to them, etc. There are tons of samples on the web. Just Google "cover letter" and "sample" and you can copy their ideas. Good luck.
