Unless you are an expert computer programmer, you really can't. You need a program that knows where to look and how to find things that are out of the ordinary, and even those programs fail a large portion of the time.
Viruses are almost never just single files that you can delete and be done with them. You'd have to look in places such as the registry, which is filled with millions of entries, all of which are encoded. Many times, a virus attaches itself to another program, and you'd have to dig through the code and remove the portion causing the problem. Worst of all, most viruses clone themselves endlessly, so even if you do find one instance of it and delete it, it'll pop up again anyway.
If you are concerned about having a virus, you can search the internet for free anti-virus software (it does exist, and is often better than the expensive software like McAfee, because people writing viruses specifically target commercial brands).

If everything you try doesn't work, then you may have to format your computer. Be sure to back everything up first, as it will ALL be permanently erased, and then format. You can find instructions for doing so in many places, but one good guide is here: http://forum.starmen.net/?t=msg&th=13142

It is extremely difficult to pinpoint which files contain a virus yourself. I would recommend using a virus-checking program, and if the program can't delete the virus, manually delete it yourself. Simply to to the directory and file listed in the program after it does the check.

Once you identify the virus, search the web, and there are
places that document viruses, and how to erradicate them.
Sometimes its as simple as deleting a file.
Some are more complex.

Tough one. It really depends on the virus but usually you would want to just delete the file that is infected and restore it from a backup. thins might not be easy due to it might be a windwos system file and can';t delete it cause it's in use,.. in that case try safemode....

Go to Avast.com and download the free anti-virus they have then let that do the job for you. Its free for home computers and works quite well

besides just deleting the infected file, you are going to have to read binary before you can ever disinfect a file.
