Sometimes windows will try to check to see if there is a disk in the drive, and if there isn't one, they can make some weird and annoying noises.

If it is doing it all the time, then the drive may be bad, if it just does it when you open 'my computer' or when some other program is verifying available drives then you can just ignore it, it's normal.

Stomachs sometimes do this too when they are empty, for the same reason. Computers become more human with every new generation! Seriously though, it's most likely that your antivirus or other protective software is doing a periodic scan and is checking to see whether the drive has a disk that needs checking.
