why password policy is disable in my PC, is it related to system file type(i.e FAT32) and how can enable?

fat 32 is kind of way you choose it while you are set up your windows and it is better than the quick format and other things and it has no relation to these type of password policy

Enable it here :
Go to Control Panel > Administrative Tools > Local Security Policy then open Account Policies > Password Policies and then change the options.
