You've received some bad answers. It was in the Fertile Crescent (modern Iran and Iraq). A good example of a very early city is Ur, from which we get the legend of Gilgamesh.

According to the bible it was Enoch, founded by Cain in the land of Nod - a bit strange as there were only four people in the world at that time.
According to Sumerian tradition it was Eridu in today's southern Iraq.
Archaeologists have also made claims for Jericho.
Cities were in existence by 4000 BC. One in Syria has recently been discovered to have been sacked and burnt at the time - the earliest evidence of war.
