Anyone can wear the number 5 shirt these days and it's becoming less fixed in terms of what position a number 5 will be, due to the squad system.
However, traditionally it has been the centre-half who has worn the number 5 shirt. A centre half usually plays in defence.
(Before 1925 a centre half usually played in what we now called a central midfield position, but that was before shirt numbers)
