Newton's law of motion states
Every object in a state of uniform motion tends to remain in that state of motion unless an external force is applied to it.
To run you need to push your body forward, which means your foot is pushing an equal force backwards. Since this backwards force is in the same direction of the earths rotation, you are technically adding to its speed. Even with everyone running at the same time, the resulting force would be insignificant compared to the force of the earth, and the results would not be measure-able, except over an extremely long time.
To slow down the earth, everyone would have to run in the direction of the rotation.
The drag of the moon and the tides are probably more noticeable in slowing down the earths rotation then all the people in the world running in one direction.

Ha, Ha,...not likely....but I read somewhere someone came up with a calculation that if everyone in China got on a 3 foot high chair and jumped off simultaneously , they could great a shock wave that would destroy the west coast of the US.
Maybe they should give it a try ...I never liked San Fransico anyway.

The mass of all people living on the planet currently is still less than 1/1,000,000,000th of one percent of the earth's mass. Any action done by even every single one of the humans on the planet won't make any difference to the earth.
Except pumping millions of tons of carbon dioxide into the atmosphere every day...*that* is how we're affecting the earth :(

No, because when the earth stops in "neutral", gravity would be lost and we would all float off into space and die.
