Boston, Chicago, and NYC are the first ones that come to mind, but you can make a name for yourself in your home town before taking your act on the road.
You have a better chance of being seen by alot of people in a big city, but a better chance to learn and grow in a smaller one. Get a bit of experience and a following in the area where you live and then bring your act on a few tours of the cities I mentioned.
There's lots of competition in the big cities so you really need to have your act together before making the jump to move somewhere more populated and further your career.

Lots of people will say NYC or L.A., but that's a good place to get lost in the shuffle.
Move to a moderate sized city, like a Tulsa or an Albany. Try your material there. If you're good, you can build a following, and then take it to a larger city.

They say Chicago is the comedy capital. So i'd go with Chicago, southern California, or NYC.
