It was shown in the background only for a minute while Jennifer Lopez was stumbling up the road.. The church was lite up in the backgound, and I think it was in San Francisco. none of the weddings in the movie took place there, it was just in the background.

Assuming that the picture in the link is the church than it is St Ignatius which is on the University of San Francisco campus.

I found a link with a picture of the church you're talking about, but I don't know the name of it. The exterior of J.Lo's home was filmed on Montgomery Street, just south of Union Street on Telegraph Hill. Maybe by using that information you will be able to do further research and find out the name. Here's the pic of the church:
