But apparently everyone here is supposing that you mean "best book ever read" and in that case currently, "Les Miserables" by Victor Hugo because it's importance of humanity and universal themes. DO NOT just watch the theatre event but read the book because of his language.

Of course this a very subjective question and I think it is hard to pick just one.
My favorite book of all time is The Phantom Tollbooth by Norton Juster. My favorite book currently is Sock by Penn Jillette. I also like Lamb by Christopher Moore quite a bit.

If you are going to speak objectively, I would have to say it would be Mary Shelly's Frankenstein, being that it is the first novel ever written. It is actually classified as a novella due to its length; however, it is the first story written in the structure that we recognize as a book today.
On a personal note, My fav is George Orwell's 1984.
