Ooh, that's tough! I'd probably have to say Chloe Sevigny in "If These Walls Could Talk 2". I've heard from tons of femmes that she is the reason they realized they love butches, and I swoon every time I see her- just the perfect blend of good looks, tough exterior attitude, and sweet gentleman. Sigh . . .
Gina Gershon from "Bound" comes close, but there's something about her smile that always bugs me; I don't know.
