How would a deletion of one nucleotide in the middle of an mRNA transcript affect the polypeptide specified by that transcript?

Any time you add or delete a nucleotide from any strand of RNA or DNA, you now have a strand that codes for something entirely different.

A deletion will result in a frame shift which will then cause the wrong polypeptide or protein to be constructed.

The other answers are correct for the most part, but there is a tiny chance that such a deletion would not affect the protein however remote. Because there are more codons than there are amino acids it is possible though extremely unlikely that a frameshift such as described by the first answer would result in nothing terrible.

the deletion of a single nucleotide would cause a frameshift mutation...all the amino acids after the mutation will be changed and the protein will not work.
