It means that if you are trying to get something, when you acquire it, you will not be as happy as you thought you would be. for example, if you think that having a lot of money is the ultimate goal, and you work all the time and sacrifice your family and personal life for money, you will find that when you have a lot of money, it does not bring the happiness you thought it would. It could also mean that the things you want a lot, the things you "chase", or work hard for, are so intrinsic to you soul, to your character, that you can't not "chase them", that you have to search for them. This could be a good thing if you are chasing the love of your life, but more than likely would be a bad thing, like an addiction.

There is always something that you will want to pursue on this earth. When speaking of earthly things after you have completed it, you will find something better. Even if you decide to stay at what you have conquered the nagging feeling of more will always be with you.
That is why the pursuit of happiness is always continual.

here's an example:
you're chasing(not literally) a boy. and no matter how hard you try, you might get him but that doesnt make your life better. good things come to you. you dont come to them. get it?

What it really means is that wanting something can be very different than having something.
