I'm assuming you don't mean someone physically opened your computer and removed memory from you system. There are many malicious programs out there that are designed to replicate themselves on your hard drive. These programs can be installed and run with or without your knowledge. As the programs continue to replicate you would notice less free space on your hard drive with no apparent cause. The best defense is to keep anti-spyware and anti-virus software up to date. It can be extremely hard to detect and remove malicious programs even with their help.

Theres two basic types of memory 1. Ram 2. Harddrive

Ram - unscrew 4 screws from the back & open the case / push the two retainers holding each Ram stick and they detach

Harddrive - unscrew 4 screws from the back & open the case / unscrew the 4 screws holding the harddrive / unplug the power connection / unplug the ribbon connection and take the drive out

If you have a laptop, there is a lid underneath or under the keyboard where the RAM is stored. A screwdriver and you have it out.
If you have a desktop, open the case and take it out of the motherboard. Very easy.
