Determining methodology means determining what kind of experiment will give you content validity. Pathological liars pose a problem in that the answer you are likely to receive from them will skew your data unless you have controls in your methodology to extract false answers.
The spouses may be more likely to answer in a truthful fashion if separated from their spouses.
Determine a statistically valid population (say 300)
determine WHAT questions you will ask.
determine what scale of measurement you intend to use on the questions (likert most likely)

If youi have a valid population size content validity will then be based on the validity of the questions.
If you assume ALL pathological liars will lie on questionaires you are making false assumptions about your population. Some will always lie, others will lie frequently, while still others may not lie at all.
These are all factors to take into consideration in your design.
My recommendation would be to get your behavioral stats or experimental psych book out and look at the sections on study methodology.

Kind of like on Cheaters I would use hidden cameras in a room and try to entrap each individual with a decoy of the opposite sex to flirt with them and see if they take the bait or remain faithful.
You could get a girl to open up honestly with another girl but obviously this would not be as effective with the guy.

Sounds like a a homework assignment. Why would you ask complete strangers of whom you have no idea of their knowledge, to do your homework or take home test question for you?
If you lack the knowledge to answer the question yourself how can you possibly judge the quality of an answer that I might give?

But, if they constantly lie, wouldn't they lie to you about their relationship?
And, even if you ask their spouse, what if they lie to you because they're liars too or from embarassment?
