Though the term "poison" can refer to nearly any substance (alcohol is a poison, water can be a poison), it usually refers to any substance that causes us harm in modest doses. Most toxins have one of only a few effects on a victim through chemical reactions (though there are exceptions) these interactions include:

depress or excite the central nervous system (CNS) to alter the state of consciousness (and a variety of problems); constriction of blood vessels (limiting oxygen to body); inhibiting nervous tissue - limiting/ceasing response (i.e. make you stop breathing); inhibit cell transport (i.e. stop cells from working); chemical burn; and Lysis (destruction of cell walls)

Any of which can be fatal (or, at least, bad) if not treated

It depends directly on the type of poison is administrated and how, it also would be really useful to specify and know weather or not it's a mixture.
For instance hemolitic substance will destroy the blood cell which will eventually wont allow the brain to get enough oxygen. Necrotic poison will actually destroy the cells surrounding the substance depending on its power in a way similar to acids and caustics, acid this this will create internal injuries in many types of tissue allow gastric secretion, pancreatic enzymes and/or colonic bacteria into the rest of the abdomen cavity eventually contaminating blood torrent and intoxicating and/or contaminating other organs.
Depending on the poison if it intoxicate the nervous system it will induce a higher activity in a way ordering the heart to beat so fast it wont actually be able to pump at all. Or either way stopping the order of breathing and/or to the heart to stop pumping. Also could happen that the centers of conscience are affected and the person is driven in to a comatose state indefinitely.
Poisoning the kidney will kill the person by not allowing him or her to get rid off her own toxic waste throughout urine

Toxicity is a function of dosage. For instance, iron in the right dosage is healthy and good for you. However, too much and your stomach will bleed. 1 geritol vitamin has enough iron to kill a 2 year old. Therefore, it is the dosage that kills, not necessarily the specific toxin.

As a sidebar, the effects that people feel when they take drugs are effects of the body being poisoned. Hypoglycemia attacks feel exactly like a very bad drunk, so I believe that being drunk enough to vomit and pass out are probably related to too much insulin. I also feel that many of the new 'diseases' are simply our bodies becoming unable to fight off the environmental poisons we are faced with today.

There are many different types of poison, each of which works in a different way. Nerve poisons shut down the activity of the nerves which leads quickly to brain death. Carbon monoxide works by fitting into hemoblogin in such a way that it cannot carry oxygen to the brain and body. Some poisons work by 'locking up' some enzyme that is needed for metabolism. It is simply impossible to go through all the possible variations.

There are many types of poison. Some types won't kill you. They might just make you sick or something. People die from certain poisons because when the poison gets into your bloodstream, it shuts down many vital organs and then you die. Poison sucks, huh.

Well, each poison is different, but they all make the body do something that ends up being harmful, or they make the body stop doing something which is essential to living.
