**spoilers** edward and alphonse elric tried to perform the forbidden human transmutation to revive thier dead mother when ed was 11 and al was 10 years old. edward lost his left leg whereas al lost his entire body. ed then binded al's soul into an armor tat was nearby and lost his right hand through the equivalent trade. their childhood friend, winry and winry's grandma (pinako) installed edward with automails in place for his lost limbs. they then went on a journey (which includes edward becoming a state alchemist so that he can access to confidential information) to find the fabled philosopher's stone in order to return to their original bodies.

This is the Preamble to the show and the 1st law of Alchemy. Edward and Alphonse Elric broke this law (although they tried to follow it) when trying to ressurect their mother who died after being ill for some time. They managed to get the equal amounts of that which the body is made of but could not balance the sould and personality. This caused a backlash of power which caused Edward to lose a leg and an arm and Alphonse to lose his body. Edward's innate talent and skill saved Alphonse by sealing his soul to some armor. Their neighbor and friend Winry, created an Auto-Mail arm and leg for Edward. Think of a cyborg and you get the idea of the arm and leg.
In order to reverse the damgae that has been done, Edward joins the State Alchemists so that he can have access to their library and other sources of information so that they can find the Phoilosephers stone which should be able to put things back to right. Due to this quest the boys grow and mature after seeing the world and are able to strengthen their bond and moral uprightness.

Their father abandoned the family (he was a humunculi If I remember correctly) and their mother died when they were both very young.
They attempted to use alchemy to bring their mother back to life and it obviously failed. That is how their physical appearences came to be what they are, through the failed atempt to bring their mother back.

The Elric brothers tried to revive their mother with alchemy,but failed.Because in alchemy human transmutation is forbidden.Edward suggested to Alphonce to revive their mother after she died.Ed was 11 years old, Al was 10 years old.They studied alchemy on their own, and performed human transmutation.Ed lost his left leg,but he saw what was in the gate.As he thought it was the "truth" it was as if almoust all knoledge about alchemy was forsed in his head.Al was taken into the gate as a sacrifice.Ed acted quickly to save his brother.Ed atached Al's soul to the armour, by sacrifising his right arm.After that Ed was equiped with Auto-Mail.He desided to become a state alchemist.Becoming one would provide all the knoledge he needed to regain their original bodyes.And.so their adventure began.

where have you watched up to? in the earlier part, one lost his arm and the other his body because they both tried to bring their mother back from the dead, there is a rule that in order to do that, something has to be exchanged, so he lost his arm and the brother, his body. they just barely managed to seal the younger brother's soul in that robot. the mother came back, but as something else, not human.
