The Vertical Assembly Building in NASA's Kennedy Space Center, formerly used for final assembly of Saturn V rockets, now for the Space Shuttle. It's the 3rd largest building in the world by volume, but the largest as a single room. It even has its own weather system.
This SINGLE room has about 130,000,000 cubic feet of space, which other contenders should check against.

i am going to give you more of an abstract answer. like said above, the room of improvment is the biggest room. but where is this room located, in the heart. in the heart, the largest room, are many many more doors, and when you go through these doors, even more doors, now imagine how big the first room has to be for there to be these many doors(try to imagine, it took me a while to understand).
