On the website I provided it has pictures of the male and female hamsters so that you can get a visual.

The female hamster has two clearly seen openings, the vagina and the anus. The vagina is just above the anus, and their is very little space between them. In adult animals, this area is surrounded by pigment. These two openings are positioned very close together.

The male hamster also has two openings, the penis and the anus. By the time the hamsters are weaned, the distance between these two openings should be clearly seen in male hamsters. Also as it comes closer to the time of weaning, the male hamster's testicles will begin to drop, giving the rear end a rounded appearance compared to that of the female.

Using the clear box trick, look for large testicles on the underside of the hamster. In males, there will also be a bigger gap between the urinary and anal openings. The perineal area (or the area between the genitals and the anus) comes to a point at the base of the tail in females, but this area looks more rounded in males.
