I have heard numbers from cast members but it's never the same number twice. I do know a lot depends on factors such as Staffing and the number of attractions open. The most common numbers I hear is between 65,000 and 80,000. Although I tend to think it is closer to 65,000 than the 80,000. At this point the Fire Marshall makes the decision to not let anyone else in.
Although there are other levels that Disneyland does do such as limitting ticket sales or restricting new entries for a few hours.
This is for Disneyland in CA. If you are asking about Walt Disney World in Florida I am not sure on that one.

28,000 of the total 48,000 can fit inside the Disneyland California Adventures addition.
