I assume you mean "Stratosphere to Mandalay Bay".
It usually takes about 30 minutes to 60 minutes. On a night with heavy traffic, it could take longer.
Don't drive it. Walk it in small chunks.

From a former resident of LV, let me tell you that any night of the week it will take you at least 30 minutes.... Friday and Saturday and sometimes Thursday nights, expect at least an hour or so if you really want to do it... most avoid it and take side streets. During the day it can take anywhere from 10 minutes to 30 minutes, depending on time of day, road construction, etc. It's several miles from one end (Mandalay Bay) to the other end (Stratosphere). One last word of advice - from your room on the strip, other resorts always seem closer than they are - if you plan on walking, you might find it a longer hike than you expected. More than one of our friends came to town and were unpleasantly surprised by that. whoops!

With traffic - an hour. (Like any Friday or Saturday evening.) Without - ten minutes.
