What message does the signalling one mean to communicate when he makes a thumb-up gesture to another -- when this is done:
1. By a man to another man?
2. By a woman to another woman?
3. By a man to a woman?
4. With a single thumb?
5. With both thumbs?

In general, it's an encouraging sign meant to tell the receiver "good job", "way to go", "all right!" I don't know that there's a lot of difference in how men use it and how women use it, but there could be. Probably the situation in which it is given lends more shades of meaning to it than who is giving it and who is receiving it.

For example, if a friend who is nervous about speaking in public gets the "thumbs up" from a friend in the audience, it would mean something like, "Don't be nervous. You're doing great." If two women friends were out together and a man approached to ask one of the women to dance, a discreet thumbs up by the other woman might mean, "Wow!" or "I'd go for it."

I would say that two thumbs up just indicates double enthusiasm.

The usual meaning is an affirmation, or a positive indicator. It can mean yes, or OK, or good, or great, or I agree.
