My favorite is in "It's a Wonderful Life" when Jimmy Stewart realizes the value of his life and has been allowed to go back to normal again, he is running down the main street in town shouting "Merry Christmas!!" to everyone he sees. Man, that just chokes me up every time I see it :)

There are so many...
There is of course
..." Use the Force Luke" from Star Wars
... Jaws " I think we are going to need a bigger boat"
... the orgasm scene from When Harry Met Sally
... the I love you speech from The American President "I need to say something here, but Im probably going to screw it up. I'm in love with you. Im sure of it. But I'm wondering if its best if I just went away for awhile." to which he answers " .... besides if you went away, Id find you".
...Dory in Finding Nemo..."just keep swimming"
...ET "Ill be right here"
a scene with little to no words in Close Encounters of a Third Kind where the aliens pick the normal guy (not the astronaut) out of the line up and pick him to go in the spaceship
The end of "One Flew over the Kookoos nest" where the huge Indian smothers Jack Nicholson's character because they did electroshock on him and he's totally lost it
Robert Redford in The Natural where he hits the baseball and it turns the nitetime stadium lights into fireworks.
In Backdraft where the Chicago Fire Department is in processional to the firefighters funeral in fulldress uniform and the bagpipes are playing
And probably one of my all time favorites
The Man From Snowy River where he rides the horse down the basically vertical drop

The final battle in The Good, the Bad and the Ugly.
Perfectly filmed. A haunting musical score by Ennio Morricone along with that atmosphere of pure tension makes it so memorable. I don't want to spoil anything, so just see it!

"quiz show" - the scene between Ralph Fiennes and his dad eating chocolate cake in the kitchen late at night. You can tell that he wants to tell his dad so much that he is cheating, but he has so much conflict he cant.
The tension is wonderfully tight, but at the same time a subtle undercurrent. A great scene from two great actors.
