A group of people (could be a group of friends, or completely anonymous) create a "football league". Most are available online. Some leagues charge an entry fee and give prize money, others are free and you only get bragging rights.
Next, on a day specified by your league commisioner, you have a draft and get players on your team. You will draft players for various positions (usually offense only) and you will draft the defense of an NFL team. Usually you will have more players than playing positions. Drafts can be automatic where you create a preference list and the best player availabel on your list is given to you when it is your turn. The draft can be realtime online where each person makes a pick in turn. The draft can also be manual where the participants gather in a room and draft players in turn. The commisioner records the draft results and enters them into the system.
Each week you decide which of your players will be playing and which players will be on the bench. The players gain points for various categories that are set up for each league (yards gained, TDs, etc). There are negative points too (fumbles, interceptions, etc).
Each person in your league has their own team and no players can be on more than one team. During the season you can usually trade players with other teams, drop players and add players that aren't on any other team in your league.
Teams are matched up against each other each week and the total points that the active (not bench) players accumulate are compared. The team with the highest point total wins. The teams with the best win/loss record advance to the playoffs (which occurs during the last 3-4 weeks of the regular season).
