2D or 3D?

2D: any decent paint program

3D: Lightwave, 3D Studio Max, Maya, etc.

It depends a lot on what kind of animations you are referring to.

Below are my personal preferences:

Vector animations: Macromedia Flash, SwishMax.
Gif animations: Adobe ImageReady, there are probably others but I use this.
3D animations: 3D Studio Max
Video Post Productions: Adobe After Effects
