For free download and usage Spybot S&D and Ad-Adware SE (by lavasoft) are the best i have used over the years. I suggest you download them both and i have found that they occasionally pick up something the other one has missed.

The real trick to using these applications is to make sure you update them regularly. This is easy to do - Ad-Adware will prompt you with a message box, where spybot you need to press and update button. I would suggest you do this weekly to keep all the applications current. Remember, spyware is a moving target.

I work in a school district--we use Spybot and AdAware, both great programs.

When I got spyware that these wouldn't take care of, I purchased Webroot's Spysweeper. Awesome program--keeps my computer spyware-free!

Ad-aware by far. It is free and not very intrusive when you don't need it to be. If you are looking for software that will constantly remind you of it's status, look for windows defender.
