If a woman is Rh negative and her partner is Rh positive, the baby will most likely be Rh postive.
During delivery, there is a chance that the mother will be exposed to the baby's blood. Since the mother does not possess the Rh factor, her body will begin to make antibodies against it. Therefore, in subsequent pregnancies, these antibodies will cross the placental barrier and attack the growing fetus.
RhoGam, also known as Rh Immune Globulin potects Rh-negative mothers against the production of these antibodies following delivery. It is a form of the antibodies that the mother is at risk for developing; however they are injected in such low quantities that they will not attack the developing fetus. Because the antibodies have been injected, the mother's body will not make them after delivery. Eventually, the injected antibodies will break down and go away, therefore causing no problems for the next child.

HI. I am blood type O- and I need RhoGam since I am pregnant and my hubby is RH positive. The first comment said it is because you are blood type O, that is not true. It is becuase you are RH Negative as you clearly seem to know. The reason for this shot is so that your blood and your partners blood do not mix thus making them "fight" (not in medical terms) RhoGam prevents the "fighting" and doesn't allow harming the baby in your womb. It is usually given at 20-22 weeks of pregnancy, and before you give birth given that the baby is born Rh positive. If the baby is born Rh negative, then you only receive the shot once.
My friend who is also Rh -, (A-) had to go through this. Her baby was born Rh - also and she only had the shot during her 21 week.

Mother- Rh-ve. Father- Rh+ve. Baby may be +ve or -ve. If the baby is -ve, no problem. If the baby is Rh + some of his RBCs will cross into the mother's blood at the time of birth. The mother will then start making antibodies to Rh antigen contained in the baby;s red blood cells. If Rhogam is given at birth, it is an antibody which will destroy the baby's red cells before the mother has a chance to produce antibodies. If the mother produces antibodies, they can cross through the placenta into the next baby and if he is Rh+ve, destroy his RBCs, producing jaundice and anemia.

The shot is given to those who have counteracting blood.
I didnt need to get it cause Im A+ and my husband is O+
I would have needed to get the shot say if I was ( - ) cause then our blood together which is infused in your unborn childs dna would start to attack one another, there for killing/harming your unborn child. The shot makes it to where those counteracting blood types can live peacefully together for 9 months until your child is born.
