i think it can be a combination of all those things,,,, and sometimes even a negative environment or upbringing can instill strength and determination in a person,,,,,

I would say that for a normal human, if environment, upbringing or education does not ruin you, then you have that existent quality.
On the other side of it, repeated failure may be due to persistent attempts for an impossible objective. That could be taken as a symptom of brain damage of birth defect.

All people are different and seek different ends for different reasons.
Beyond that, no, I don't believe there is a genetic pre-disposition responsable for the drive to succeed, beyond the most basic desires and vauge instincts. Genetics is simply not ever that specific.
Also, I am fairly certian being a disoriented idiot is not a violation of Y!Q&A Community Guidelines or TOS. ;)

it is all of the above...education and upbringing often instill values within an individual that motivate them to continue striving and these qualities are LEARNED..determination is picked up and notice that these individuals eventually do accomplish their goal if they try enough times...thats why failure exists..because we dont try enough times not because one attempt was unsuccessul...when we give up is when we truly have failed.
