Definitely Rome for me. I have been to all three and I like Rome the best. Barcelona is okay but nothing I saw there really thrilled me and some of the people were a little rude. Venice is interesting to see once and then leave. It is not a fun place to stay. Rome is awesome. There is so much cool stuff to see and do. The old Roman ruins such as the Forum and Colusium are amazing. The Vatican musuem has some really interesting stuff. Rome is also fun just to wander around and look in all the neat stores. There are many beautiful parks and churches everywhere. It you are going for a week then you can take a train to Venice over night if you just want to see it. Florance and Pisa are also day trips from Rome.
Have a good trip.

I've travelled in both Spain and Italy. And for the western tourist, if you don't speak the language, I'd take a tour with a group -- it's easier, you have help with language or emergency issues, you get to see great sights and not worry too much about the details, like where to eat, where to sleep, & where's the bathroom. I took a tour in Italy and saw both Rome and Venice, so I enjoyed each because I had fewer worries, but my vote would go to Venice -- an incredibly unique and beautiful city. Getting lost on the streets and canals of Venice is like living in a dream world of beauty and adventure. Rome is much more like any other big city (crowds, crime, pollution, etc.), but with fantastic architecture & museums.
In Spain, however, I was an exchange student, so I spoke the language (sort of) and lived with a family. I didn't encounter the rudeness that previous postings mentioned because I wasn't a "tourist", exactly, but I did encounter some anti-American sentiment. (I saw none of that in Italy.) Again, though, Spain is much easier to see with people who know what to see. I would definitely take a tour. Spain's beaches, however, esp. farther south than Barcelona, I think are better than anything I saw in Italy.
All in all -- my pick would be Venice.
Have fun!

Venice is impressive, but you don't need more than 2-3 days to visit.
Barcelona and Rome are big cities with lots of things to do and see, so you need at least one week. Okay, if you really like museums and ancien art maybe you should need one month in Rome!
They are very different. Barcelona is modern and chic. Rome is monumental but chaotic.
If you are not too cultural, the best is Barcelona, which offers you more variety (great shopping, beaches, fashion bars and discos, and also great architecture and museums).
But Rome is a paradise if you like art and history, I love it! The only problem is transportation, as the underground system is very limited compared to Barcelona's. and u need to take crowded buses.

I went to Barcelona 2 summers ago - great architecture, great food, beach is fun, and plenty of culture. Weather should be pretty mild by April as well.
I've also been to Rome, but not Venice - so between Rome and Barcelona, Barcelona gets my nod.
